Sequencing of serum exosomal circRNAs in colorectal cancer

Data Processing :
Paired-end reads were harvested from Illumina HiSeq 4000 sequencer, and were quality controlled by Q30. After 3’ adaptor-trimming and low quality reads removing by cutadapt software (v1.9.3). The reads were aligned to the reference genome (UCSC HG19) with STAR software and circRNAs were detected and annotated with DCC software. circBase database and circ2Trait disease database were used to annotate the identified circRNAs. The junction read counts were normalized and differentially expressed circRNAs were determined using edgeR package of R software.
Data Summary :
To assess the serum exosomal circular RNAs (circRNAs) expression profile and explore the potential functions in human colorectal cancer (CRC).The differentially expressed circRNAs profiles of CRC (n=5) and age-matched healthy controls(n=5) were analyzed using RNA-seq profiling. Quantitative real-time PCR was used to validate the expression pattern of circRNAs.Bioinformatic tools including network analysis, Gene ontology, and KEGG pathway analysis were utilized.
Overall design :
Sequence was performed on 5 colorectal cancer samples and 5 healthy control samples,each sequencing sample was a mixture of 10 serum from participant.
