Small non-coding RNA profiling of urine exosomal total RNA from patients with or without prostate cancer were performed using Affymetrix GeneChip miRNA 4.0 to identify small non-coding RNA profile that can be used for prostate cancer diagnosis.
Overall design :
Changes in small non-coding RNA expression in prostate cancer patients were deteremined by comparing the small non-coding RNA expression to the positive or negative results of core-needle biopsy of patients suspicious of prostate cancer.

Biotinylated miRNA were prepared according to the standard Affymetrix FlashTag HSR protocol from 100 ng total RNA
Hybridization Protocol :
Following labeling, 21.5uL of biotin-labeled samples were hybridized for 16 hr at 48C on GeneChip miRNA4.0 Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.
Scan Protocol :
GeneChips were scanned using Affymetrix Scanner 3000 7G with AGCC software.
