Data information

Data Id :
GSE130654 ( miRNA )
Disease :
gastric cancer ( gastric )
Title :
Combination of four serum exosomal miRNAs as novel diagnostic biomarkers for early-stage gastric cancer
Percentage of samples :
Cancer : 36 / Control : 12
Data Processing :
Reads Per Kilobase of exon per Megabase of library size (RPKM) were calculated using a protocol from Chepelev et al., Nucleic Acids Research, 2009. In short, exons from all isoforms of a gene were merged to create one meta-transcript. The number of reads falling in the exons of this meta-transcript were counted and normalized by the size of the meta-transcript and by the size of the library. Genome_build: mm8
Data Summary :
Using next-generation sequencing (NGS) and bioinformatics, we identified candidate serum exosomal miRNA markers for early detection of GC in patients. The candidates were further validated by qRT-PCR in 50 newly recruited early-stage GC patients and matched healthy individuals.
Overall design :
Serum exosomal miRNA profiles of early gastric cancer patients were generated by deep sequencing, in triplicate, using Illumina Hiseq 2500/2000 platform.

Extraction Protocol :
Exosomes were isolated using the ExoQuick exosome precipitation solution (System Biosciences, Mountain View, CA, USA) according to the manufacturer’s instructions with slight modifications.RNA of exosomes was isolated using miRNeasy micro kit (Qiagen, Valencia, CA, USA) according to the manufacturer’s protocol. The extracted RNA was eluted with 14 μL of RNase-free water.
